No, they are extra cards you can hold. You can mix Special Assignment cards with Permanent ones in the same Assignment phase, so you can send more Characters to a Location than the base cards allow; actions are still resolved one by one in Initiative order, so you risk being blocked from a Location before all your Characters act.
