Can the number of players change in the middle of a campaign?

Treat it as fixed. The forum advice is not to change it, since setup depends on the count (the rulebook has you remove 3-player and 2-player buildings) and later cards may refer to the number of players. The rulebook itself does not forbid it explicitly.
