No — Expert Handling is itself an action that grants a barrel roll, and a ship can't do the same action twice in one round, so it can't follow a normal barrel roll or be combined with Vader's two actions to roll twice. If the roll can't be performed, the rest of the card's effect, including removing an enemy lock, doesn't happen; the FAQ quoted by repliers allows checking beforehand. Vader can use Expert Handling as one of his two actions together with a different action.
