A normal melee attack needs adjacency, and two spaces separated by a door are not adjacent, so you cannot melee a figure through a door. A door itself can be attacked by a figure in one of the spaces sharing an edge with it, or by a Reach attacker in or adjacent to one of those spaces who has line of sight. Missions may override this.
