Does a Mobster's Mob War strength modifier apply when I take over its Speakeasy or when my Associate defends?

No. The +1 or +2 modifier applies only to the Mobster's own attack in a Mob War. To take over one of its Speakeasies you must beat the Strength tile's base value, and an Associate you pay adds just its base strength when defending.
