Can I destroy one of my people to play a new one while I still have empty spaces?
The rulebook's v1.2 wording applies only when you have no space in any column; a publisher reply and a player's reading agree.

No — the destroy-to-make-room option exists only when none of your columns has an open space. If any space is open, you play the person there instead.
