After moving into a Room that is empty of Characters and Intruders, whether by Movement, a Distraction-style card, a Vent or Cave move, or an Escape. You also roll when a Bag Development draw or an event calls for it. No roll if anyone is already in the Room (Eggs and Germinators do not count as someone), when exiting an Elevator, or when you use a Room Action without entering the Room.
