Do I need the leaders before I launch to the Development Office?
The rulebook says you can build on any planet where you have a transport and recruit leaders at any time on your turn.

No — the leaders only need to be on your station when you perform the develop action, so you can launch there, recruit a leader from a region where you have ships, and then build. You develop on a planet where you have a transport, which need not be the active region.
