Can I defend with Shields if I spent all my Armed Markers on my own turn?

No. Markers are only re-armed at the end of your turn or in the Status phase, so Markers spent on engines or weapons are not available for shields. Spending all four on your turn leaves nothing to defend with, even against asteroid damage, because shields must be actively powered like any outfit.
