Must I follow suit with a submarine, and can I play a submarine when I lack the led colour?
The rulebook states submarines are trump, may only be played on a colour trick when you cannot follow, and must be followed when led.

Yes to both. If a colour is led and you hold it you must play it; only without that colour may you play any card, including a submarine, which then wins. If a submarine is led, you must play a submarine if you have one.
