Do one-use tool cards add to my permanent tools or count at final scoring?

No. Cards showing a one-time tool boost are kept beside your board, used once on a roll, then placed face down; they never count as tool value in scoring. Only a +1 Tool card that adds a permanent tile counts, through that tile.
