Can one player occupy several Civilization cards or Building tiles in a round?
The rulebook says each card and building holds one figure, and the per-location rule is about the same location.

Yes, one per turn. Each card and tile is its own location, so on later turns you can take more of them if they are still free, provided you do not already have figures on that spot.
