The designer said lighting does not change the effect.

No. Any exposed glyph facing your home kingdom applies whether or not it glows. Lights are for visibility only. An exposed but unlit glyph usually means a wrong seal was removed earlier; do not undo it, play on, and treat any exposed glyph as active. Glyphs hidden behind a seal do not apply.
