In a 2-player game, do the ownership markers placed on villages 6, 7 and 8 matter?

Yes. They are real ownership markers. They exist so the nations already own one location each at the start, which makes cards that need e.g. three locations of a nation reachable in a 2-player game. They can be displaced by combat like any other marker.
