No, not at once. A fulfilled quest stays gone, and explorer-track quest spaces are never refilled (explorers skip the empty space). Quests that nobody takes stay on the board into later rounds. New quest tiles come only from the setup or story card, which adds them between rounds, and a few legacy tiles such as the Puerto Cabezas city quest are refilled when claimed.
