Whoever next assigns an agent to that exact space collects them as the first step of the action, and can spend them within the same action. In a multi-space building such as Waterdeep Harbor or Cliffwatch Inn, resources belong to the specific space they were placed on, not the whole building. Resources can be added to spaces already holding an agent, but that earlier agent does not collect them.
