Can I play a unique ally again after the first copy was destroyed?
The rulebook says a unique card may enter play while another copy sits in a discard pile.

Yes. Uniqueness only looks at cards in play, so once the first copy is in the discard pile you can play another. There is no separate dead pile that blocks it.
