Do Staff cards keyed to a die number (Bootblack, Checker, Laundress and so on) trigger when I use a 6 die as that action?
The designer and publisher both ruled on this for the Bootblack, and players apply it to similar cards.

No. Those cards trigger only when you actually take a die showing that number. A 6 die used as a 4 or 5 does not count, and the 6 die still goes on your Turn Order tile.
