Can I dismiss a persistent card and then use it to negate damage?

Yes, as long as the card has no Lost icon. Dismissing it first moves it to its destination, then you can use it to negate damage. A B-side persistent card lands in the discard pile and can be lost from there. An A-side one flips to B in your hand, where it counts as a B-side card, so you can discard it with a second B-side card. A card with a Lost icon goes to the lost pile and cannot be used this way.
