How does a Two-Color card count for the no-same-colour-neighbour rule?

Both of its colours count: it cannot touch a card of the same colour on either side. On rows that need a colour sequence, such as Lake or Swamp, it may count as either colour. You may orient the card as you wish when you play it, but it cannot be rotated afterwards.
